West Yorkshire Police officers were called to Edale Avenue at about 00:20 BST on Saturday after reports of a fight involving several people.
Two 16-year-old boys and one 18-year-old man were taken to hospital and arrested on suspicion of affray.
Police said one 16-year-old boy suffered a serious hand injury, while the other two were treated for non-life-threatening injuries.
Officers are continuing to carry out investigation work on Edale Avenue and Falcon Street, the force said.
Det Insp Richard Grace said: "This is a very serious incident involving teenagers on the streets of Huddersfield.
"We understand there was a house party that was attended by around 100 people, following which an altercation took place involving a number of individuals.
"We are keen to speak to anyone who was at the party, witnessed the incident, or has mobile phone footage of the fight taking place."
